My short answer guess would be "no". I'm sure that someone will offer up an in-depth explanation, but my understanding in compression tests I've done, is I've always measured the compression in the cylinder when it was firing. Without carbs, you have no way of getting the fuel/air mixture into the cylinders to fire.

I'm not sure if you can do one manually.

Yes you can do a compression test with the carbs off, most compression tests are done with carbs at wide open throttle, you don't want a restriction on the intake side screwing up the results.Google search it you'll see

I always do it with out the plugs in the other cylinders as well so the motor turns over better, remember your checking the compression ( wall, ring & valve condition ) as it cycles thru its rotation,

Just to clarify - my "no" was only a guess ... I've always done it with the engine assembled - sorry to mislead.
